Indonesia has a high biomass potential that can be used (Riyanti 2008). Indonesia is a country with high agricultural yields. However, the high yield of agriculture in Indonesia is directly proportional to the agricultural waste produced. One of the agricultural waste can be rice straw. The selection of agricultural waste in rice straw because rice waste is abundant but has a low economic value. This study aims at determining the modifications of the components of the rice straw fiber after a preliminary biological and chemical treatment, in the first step of the bioethanol production. The analysis of the components of the rice straw fiber was carried out in several steps, namely the preparation of the samples, the biologically pretreatment process using an efficient microorganism (EM4) while chemically using sodium hypochlorite (NaOCl ), then testing the levels of hemicellulose, cellulose and lignin by the method of Chesson, 1981). The results showed that biological pretreatment reduced lignin levels better than chemical pretreatment. but for hemicellulose and cellulose levels, each pretreatment has increased. The results of the study suggest that pretreatment, both biologically and chemically, can alter the fibrous component of rice straw.
