Analisis Keragaman Genetik Tanaman Pala (Myristica sp.) di Minahasa Utara Menggunakan Penanda Molekuler Random Amplified Polymorphic DNA (RAPD)

Search results based on morphology of nutmeg plant there are several types of nutmeg plants that grow in North Sulawesi, especially in North Minahasa Regency. Previous research on nutmeg plants has been done a lot but information about the genetic diversity of nutmeg plants is still lacking, especially in North Minahasa regency is still minimal. This study aims to determine the genetic diversity of nutmeg (Myristica sp.) In North Minahasa based on the Random Amplified Polymorphic DNA (RAPD) molecular marker. Analysis of genetic diversity of nutmeg plants was carried out through several stages, namely DNA extraction and purification, DNA concentration and purity testing, target gene amplification using PCR (Polymerase Chain Reaction) method and finally visualization of amplification results with 1% agarose gel electrophoresis. Data analysis using IBM SPSS Ver. 20.0. The results of the study using six primers RAPD OPT 07, OPT 16, OPH 06, OPA 18, OPH 01 and OPH 02 showed that North Minahasa regency had a diverse population of nutmeg plants as indicated by the formation of polymorphic bands / samples of nutmeg used. Based on the analysis of dendrogram using SPSS 20 application shows that six samples used from three locations in North Minahasa consist of two different groups, generally based on clusters formed from the six primers used can be said to show polymorphism or variety.
