Thermophilic bacteria have economic value because they produce thermostable enzymes useful for the industry. Linow Lake is a volcanic site located in the town of Tomohon, in northern Sulawesi. The research was conducted at the Microbiology Laboratory of the Department of Biology of FMIPA Manado State University from April to September 2019. This study aims to obtain isolates of thermophilic bacteria and proteolytic activity. This research uses descriptive qualitative research methods. The data obtained were presented in a descriptive way, covering macroscopic characteristics and qualitative qualitative tests. The proteolytic test was performed qualitatively by measuring the diameter of the clear zone created around the colony. The stages of the research were bacterial isolation, the thermo-tolerance test and the bacterial proteolytic test. According to the results of the study, we obtained 18 bacterial isolates capable of developing in the temperature range between 45°C and 60°C. It has a proteolytic capacity by producing clear areas on media skimmed milk agar. The results of the qualitative test of the proteolytic activity of 18 isolates, two isolates with the largest clear-zone diameter, were DLII.13 isolates with a qualitative activity of 6 mm and DLI.2 isolates, of 5 mm.
